Secondary antibodies are monoclonal or polyclonal antibodies bind to primary antibodies or antibody fragments. Secondary antibodies provide signal detection and amplification along with extending the utility of an antibody through conjugation to proteins. Secondary antibodies can be conjugated to enzymes, fluorescent dyes, or other molecules, which include but not limited to horseradish peroxidase (HRP), alkaline phosphatase (AP), fluorescein isothiocyanate (FITC), rhodamine derivatives, Alexa Fluor dyes. In biological research, secondary antibodies have been widely used in various biochemical assays, such as Western blot, immunostaining, immunohistochemistry, immunocytochemistry.

Compared with the direct detection using primary antibodies, the indirect detection of secondary antibodies presents increased sensitivity, diversity, flexibility, and versatility. In addition, secondary antibodies bind to the heavy chains of primary antibodies so that don’t interfere with the binding between primary antibodies and antigens.

Recombinant proteins are frequently expressed along with a tag epitope, a small antigenic sequence recognized specifically by anti-tag antibodies.
