Anti-Glycan Antibodies in Detection and Discovery of Glycoantigens

Glycoantigens or carbohydrate antigens, actually are polysaccharides and glycoconjugates which can induce the production of corresponding anti-glycan antibodies and react with these antibodies. Glycan determinants are oligosaccharide domains of glycans or glycoconjugates, which play a determined action in the binding and reaction between glycoantigens and antibodies. It is estimated that there are over 7000 glycan determinants in the mammalian, of which mostly are remains unknown. Furthermore, increasing investigations have confirmed that glycoantigens are closely related to plenty of pathogenic processes, cancers and other human diseases. Therefore, it is necessary and imperative to identify and characterize glycoantigens to better understand and elucidate the mechanism of their action and to find better therapies.

In reality, it is extremely difficult to study glycans and glycoantigens since that:

• All carbohydrates and glycoantigens are not primary products of gene expression, which makes it impossible to research by genetic methods like selectively knock-in and knock-out.
• Structures of carbohydrates and glycoantigens are very complex and variable, as well as heterogeneous, which makes identification and distinguish more difficult.
• Existing biological techniques and methods are mainly for detecting and monitoring proteins which are not enough suitable for carbohydrates.

Despite these challenges, detection and discovery of glycoantigens can be carried out by using antibody-based methods and techniques as ELISA, Western Blotting, immunohistochemical staining, etc. Anti-glycan antibodies with high affinity and specificity have been being increasingly utilized as reagents in biochemical and biological researches of glycans and glycoantigens.
