Both glycosyltransferases and glycosidases are critical carbohydrate-processing enzymes involved in a number of important biological processes, such as basic formation and hydrolysis of carbohydrates, posttranslational modification of glycoproteins, and synthesis of glycosides and glycoconjugates. For example, assay α1-3 fucosyltransferases by means of capturing of the glycosyltransferase on immobilized antibodies to antigens, or binding the anti-glycan antibodies to the immobilized fucosylated glycosyltransferase in an ELISA-type format. The detection also can be conducted by using beads containing the acceptor glycan in flow cytometry that is a modification to a new structure that can be recognized by an anti-glycan antibody. While glycosidases tests can be performed by measuring the gain in the binding of anti-glycan antibodies.
