Property of Neospora Caninum

Neospora caninum is an intracellular Apicomplexan protozoan parasite. It is a causative agent of neosporosis which is a serious disease of cattle and dogs worldwide and appears to a major cause of abortion in cattle. N. caninum was misidentified as Toxoplasma gondii before 1988 because its structure and life cycle is similar to T. gondii. The life cycle of N. caninum is characterized as a complex facultative heteroxenous. It includes a definitive canid host with the sexually reproductive stage occurring, and a range of intermediate hosts with asexual replication occurring. If naïve cattle are infected by oocysts shed from definitive hosts, sporozoites in the oocyst differentiate to tachyzoites and spread through the body. Parasites can exist over long periods as quiescent tissue cysts contained within the host tissue.

Neosporosis is a serious infectious disease of animals. however, so far, no efficient vaccine has been developed. Monitoring the N. caninum parasites to reduce the likelihood of infection is an urgent issue for protection. For diagnosis of neosporosis, various methods have been developed, such as IFAT, immunoblotting and ELISA. Several targets have been identified for the development of detective reagents:

  • NcSAG1
  • The surface-associated protein 1 of N. caninum (NcSAG1) is expressed in the tachyzoite and downregulated during the conversion from tachyzoite to bradyzoite. NcSAG1 has been identified as one of major surface antigens of N. caninum tachyzoites and demonstrated to be immune dominant and involved in interactions between the tachyzoite and the host cell.

  • NcGRA7
  • NcGRA7 protein is an immunodominant antigen shared by both tachyzoites and bradyzoites. The titers of anti-NcGRA7 antibody in cows with a history of abortion is significantly higher than in non-aborting cows infected with N. caninum. In addition, the frequency of anti-NcGRA7 antibody-positive individuals was higher among cows with a recent history of N. caninum-associated abortion than among cows that had experienced N. caninum-associated abortion more than 30 days previously.

  • NcSRS2
  • There are also many surface proteins structurally related to NcSAG1, which are designated NcSAG1-related sequences (NcSRSes). NcSRS2, with a molecular weight of 37 kDa, elicits strong antibody response in infected animals and is an attractive candidate for diagnosis and vaccine antigen. In vitro studies have shown that blocking NcSRS2 can limit the parasite's ability to attach to and invade host cells. In addition, the recombinant NcSRS2 (rNcSRS2) exerts a protective effect against encephalitis and transplacental transmission in in vivo studies.
