Clinical Significance of Antibody/Peptide Targeting Strongyloides

Strongyloides is a genus of parasitic nematodes that are found throughout the world. This genus infects a wide variety of vertebrates such as mammals, birds, amphibians and reptiles. Two species (S. stercoralis and S. fuelleborni) infect humans, resulting in intestinal infection called strongyloidiasis. S. stercoralis is the most common and globally distributed human pathogen of clinical importance while S fuelleborni is found sporadically in Africa and Papua New Guinea. Because the parasite load is low and the larval output is irregular, so conventional diagnosing strongyloidiasis is particularly challenging. Recent studies have shown serological diagnosis is an important alternative approach for supportive diagnosis in acute and early cases of strongylodiasis. The detection of the host antibodies against Strongyloides antigen is a major method for serodiagnosis of strongylodiasis, thus making it essential to discover Strongyloides-specific antigens.

Phage-fused Peptides for Serodiagnosis of Strongyloidiasis

Phage display is one of the powerful tools used for the discovery of antibody and peptide targeting parasite. Based on this technique, Nágilla et al. have identified five peptides that mimic S. stercoralis antigens and have a strong immunoreactivity to circulating IgG from patients' sera with strongyloidiasis. More importantly, two of them exerted the good diagnostic accuracy, with high sensitivity and good specificity. This suggests selected phage-fused peptides can be referenced as mimotopes and used to detect circulating IgGs during Strongyloides infections.
