Catalytic antibody (abzyme) is a type of monoclonal antibody with catalytic activity. Natural abzymes can be found not only in healthy humans and but also in patients with autoimmune diseases such as systemic lupus erythematosus. Anti-vasoactive intestinal peptide autoantibodies are found in normal humans, where they can bind to and hydrolyze DNA. Abzymes are usually generated by the immunization of animals with synthetic haptens. So far, this technique has proven most successful for the elicitation of abzymes that approach the catalytic efficiencies of natural enzymes. Phage display is a powerful and widely used technique in the biological and medical research fields. The rapid recovery of specific antibody candidates by robust screening of a variety of target antigens is one of the most successful uses of this technique today. Using a suicide substrate of β-lactamase as a target, catalytic antibodies with β-lactamase activity have been identified by phage display technology. Studying abzymes can provide important insights into enzyme reaction mechanisms and the immune system itself.
