Introduction of Whole Cell Immunization
Whole cell immunization is a novel strategy that the antigen overexpression cell line can be used as the immunogen. Compared with the normal immunization strategies, this strategy can benefit from higher immunogenicity, naïve antigen conformation and avoid the complex purification process, which is specifically suitable for novel antibody discovery against membrane protein (extracellular domain). Besides, this strategy is also a suitable method to identify new biomarkers from the surface proteins of tumor cell lines, thus can contribute to the cancer diagnosis and also the development of novel therapeutic reagents.
Applications of Whole Cell Immunization
The whole cell immunization has been widely used for the antibodies generation. Compared with other immunization strategies, whole cell immunization has been successfully used for the mAb generation against cell-surface proteins as membrane and membrane-associated protein. These cell-surface proteins may lose their native conformation during the regular purification process and result in the challenges for discovering specific mAb against the conformational epitope. By generating the special cell lines that overexpress the target proteins, their natural conformation and also the modifications can be maintained, our scientist can therefore lay a good foundation for the high-quality antibody discovery at the early stage of animal immunization. After screening using the target cells and counter-screening using negative cells (the same cell line but do not express the target protein), the target-specific binders can be selected for further validation. Creative Biolabs can employ either hybridoma, phage display or other novel techniques for the selection and validation of specific monoclonal antibody. In addition, we can also use this strategy to discovery single domain antibodies with high affinity and stability.
