Sunflower trypsin inhibitor (SFTI), a potent inhibitor of trypsin, is a small, ribosomally synthesized circular peptide of 14 amino acid isolated from the seeds of the sunflower Helianthus annuus. SFTI is the smallest known member of the Bowman-Birk family of serine protease inhibitors which interact with the enzymes they block via an exposed surface loop adopting the canonical protease inhibitor conformation. As a result, the noncovalent complex inactivates the protease. Another particular feature of the Bowman-Birk family proteins is the particularly well-defined bonding loop which is a short β-sheet region linked by disulfide bond.

SFTI scaffold can be a promising candidate for peptide-based drug design. As a small size peptide, SFTI comprises two short antiparallel β-strands connected at the reactive site end by an extended loop region, and by a hairpin turn at the opposite end. Its stability based on a cyclic backbone, a network of hydrogen bonds, a structure constrained by three proline residues and an intramolecular disulfide bridge for additional rigidity. SFTI is amenable to chemical synthesis, allowing for the creation of synthetic variants. The creation of linear variants of SFTI offers an option for studying the putative linear precursor peptides. In addition, SFTI may be considered as a possible scaffold structure for the future development of peptide-based radiopharmaceuticals.
