Drug-resistant Bacterial Infection

Due to the limited treatment options available and the historically slow pace of development of new antimicrobial agents, the emerging and re-emerging of multidrug-resistant (MDR) bacterial infections in the communities and health-care settings constitutes a serious threat to global public health. Infections due to MDR strains are associated with increased morbidity, mortality and prolonged hospitalization, which translates to a significant burden on healthcare systems. The the most frequently isolated MDR bacteria are Enterococcus faecium, Staphylococcus aureus, Klebsiella pneumoniae, Acinetobacter baumannii, Pseudomonas aeruginosa and Enterobacter. Antibiotic-resistant bacterial increases in number as time goes on. From an evolutionary perspective, bacteria has two major genetic strategies to adapt to the antibiotic attack, gene(s) mutations and acquisition of foreign DNA coding for resistance determinants through horizontal gene transfer (HGT).

Antibody Targeting Bacterial Surface Protein

The traditional antimicrobials are declining in efficacy and no longer work well as they used to be. Because of playing vital roles in immune responses, antibodies have been successfully exploited to treat bacterial infections and cancers. For bacterial infections, the whole molecule (IgG) format or fragments can bind specifically to the antigen and block the interaction of the latter with a conjugate ligand. The combination of antibody specificity and killing effects of antibiotics meet the need for novel therapeutic strategies, thus providing the great chance for clinical success.

In general, immunological therapy approach against bacterial pathogens involve antibodies directed against cell surface-exposed virulence-associated epitopes or toxins. Surface proteins are preferable to toxin due to its localization on the bacterial. Many bacterial surface proteins play different roles in innate and adaptive immune responses. It has been shown that antibodies against certain surface protein are beneficial for defensing against bacterial infection. In Creative Biolabs, many antigens have been screened as candidate targets for monoclonal antibody generating and the targets should possess three criteria, the targets should be highly abundant and highly conserved on bacterial in vitro and during infection, as well as absent from human cells.

Antibody-antibiotic Conjugate (AAC)

By combining key attributes of antibody and antibiotic, AAC has been shown to be efficacious against drug-resistant bacterial infection. An AAC has three components: an antibody to target delivery of the payload to bacteria, an antibiotic payload to kill bacteria, and a linker attaching the payload to the antibody. Antibodies we made could be conjugated with antibiotic through a special linker, thus forming an AAC. All three components are essential and playing different roles in an AAC. Antibody delivers the antibiotic payload to bacteria specifically and linker is involved in releasing payload under special circumstances, while the payload is responsible for the mechanism of action.
