Multiple myeloma (MM) is a hematologic malignancy that is characterized by the accumulation of abnormal plasma cells (PCs) in the bone marrow (BM). This paper details the development and assessment of AMG 701—a half-life extended anti-BCMA (B-cell maturation antigen) bispecific T cell engager (BiTE) molecule for treating MM. Preclinical studies detailed the potency of AMG 701 in inducing T-cell-mediated cytotoxicity against MM cells, pharmacokinetics, and pharmacodynamics in cynomolgus monkeys and efficiency in mouse xenograft models of MM. The study brings out the molecule's highlighting ability to deplete the plasma cells and further suggesting its potential in clinical development in the treatment of MM. It goes further to report on the interaction of AMG 701 with PD-1 blockade, along with the possibility of increasing therapeutic efficacy.
