Anti-Robo4 Antibody Development

Robo4

Robo4, which is expressed exclusively in endothelial cells and has been implicated in endothelial cell migration and angiogenesis, belongs to a family of four Roundabout receptors. Higher Robo4 expression is closely associated with distinct clinical and biologic characteristics in different tumors, such as breast, lung, colorectal cancer, and so forth. Compared with the other Robo1-3, which are mainly expressed in the nervous system, Robo4 is not only expressed in a different tissue environment but is also structurally divergent and contains only two IgG-like domains and two FNIII repeats in the extracellular domain (ECD) and lacks the CC1 and CC3 motifs found in the intracellular domain of other Robo1-3. Robo4 acts as a receptor that modulates vascular endothelial growth factor (VEGF) A-VEGF receptor (VEGFR) signaling. Robo4 is a potential tumor angiogenesis marker because angiogenesis is only activated in tumors in the adult, with the exception of some pathological states.

Immunotherapeutic Approaches

Tumor endothelial-specific expression of Robo4 in adults identifies this plasma membrane protein as an anti-cancer target for immunotherapeutic approaches, in particular, vaccines and antibody drug conjugates (ADCs). Anti-Robo4 mAbs could accumulate in the tumor without distributing to normal tissues and mAbs with high internalization activity have greater tumor-targeting potency. The first in vivo therapeutic application with anti-Robo4 mAb revealed that tumor accumulation and anti-tumor effects of mAbs and corresponding ADC with high cell-internalizing activity was significantly higher and stronger than that of mAbs and corresponding ADC with low cell-internalizing activity, respectively.

Therefore, Robo4 can be an effective marker for tumor vascular targeting.
