CD3-epsilon polypeptide, also known as CD3E,is encoded by the CD3E gene which resides on chromosome in humans. CD3E is a T-cell surface single-pass type I membrane glycoprotein and is one member of the T-cell receptor-CD3 complex. This CD3 complex also contains CD3-gamma, -delta and -zeta, and the T-cell receptor alpha/beta and gamma/delta heterodimers. Besides, the genes encoding the epsilon, gamma and delta polypeptides are located in the same cluster on chromosome 11. T-cell receptor-CD3 complex serves an essential role in coupling antigen recognition to a variety of intracellular signal-transduction pathways.
CD3E is comprised of 1 Ig-like (immunoglobulin-like) domain and 1 ITAM domain. It acts a crucial part in T-cell development, and defects in CD3E gene result in severe immunodeficiency. Moreover, it has reported that CD3E is associated with a susceptibility to type I diabetes in women. And, it has been exhibited to interact with TOP2B, CD3EAP and NCK2. In addition, CD3E is expressed in a variety of cancer cells, including endometrial cancer, renal cancer, melanoma, head and neck cancer, cervical cancer and breast cancer.
