FXYD5

FXYD5 (FXYD domain containing ion transport regulator 5) is a cell membrane localized ion transport regulator that is also known as, RIC, IWU1, KCT1, OIT2, DYSAD, HSPC113, and PRO6241. It is a glycoprotein bearing O-glycosylation and it is mainly expressed in endothelial cell, T lymphocytes, and basal cell of squamous epithelium.

FXYD5 is proposed to function in the upregulation of chemokine production. It is also involved in E-cadherin down-regulation, which subsequently results in a reduced cell adhesion. FXYD5 has been demonstrated to promote cancer invasion and metastasis, and a reversed correlation between FXYD5 expression and E-cadherin expression has been identified in many cancers. Thus, FXYD5 has been associated with cancer prognosis and serves as a target for various cancer treatments, including head and neck squamous cell carcinoma, esophageal carcinoma, gastric carcinoma, colorectal carcinoma, melanoma…. In the meantime, FXYD5 has also been exploited as a diagnostic marker for epithelioid sarcomas and it helps to distinguish epithelioid sarcomas from malignant rabdoid tumors.
