TNFRSF9

TNFRSF9 is abbreviated for TNF receptor superfamily member 9. It is expressed by antigen-presenting cells and various carcinoma cells. TNFRSF9 contributes to the clonal expansion, survival, and development of T cells. TNFRSF9 induces proliferation in peripheral monocytes and enhances T cell apoptosis. In addition, it promote Th1 cell responses by regulating CD28 co-stimulation. The expression of this receptor is induced by lymphocyte activation. TRAF adaptor proteins are reported to bind to TNFRSF9 and transduce the signals leading to activation of NF-kappa B.
TNFRSF9 is also referred to as 4-1BB or CD137. Numerous studies have established that signals via 4-1BB are costimulatory in nature. Further analysis revealed that although both CD4+ and CD8+ T cells express 4-1BB at comparable levels, upon activation, signals through 4-1BB are more biased toward CD8+ T cells, both in vitro, and in vivo.
A number of reports have revealed that TNFRSF9 is also expressed on a wide range of tumor cells, including SPC-A-1, H446, H460, and H1299. Expression of TNFRSF9 in cancer patients was found also on tumor vessel walls, on the endothelial wall, on vascular smooth muscles, and in liver tissue.
