Endotoxin Removal Kits

Introduction Workflow Our Products Contact Us FAQs

Introduction

Endotoxins are integral components of the outer membrane of gram-negative bacteria. Chemically, these molecules consist of a hydrophilic polysaccharide chain and a hydrophobic anchor known as Lipid A, which is the primary driver of biological toxicity. Because bacteria can thrive in nutrient-poor environments like water and buffers, endotoxins are ubiquitous in bioprocessing. In aqueous solutions, endotoxins self-assemble into large aggregates with molecular weights often exceeding 1000 kDa. However, the presence of proteins can dissociate these into monomers of 10-20 kDa, which are harder to remove due to their similar size to many target proteins.

Endotoxin removal is the critical process of selectively sequestering these molecules to prevent severe pyrogenic reactions, systemic inflammatory response syndrome (SIRS), and inaccurate experimental data. Creative Biolabs specialized kits are engineered to break the masking effect where LPS binds to your target product.

Workflow of the Endotoxin Removal Kit

The workflow for an endotoxin removal kit is engineered to prioritize speed, high target recovery and maximum potency preservation, typically completing the entire process in under 60 minutes. Unlike traditional methods that may take hours, this streamlined approach ensures that high-value biological assets are purified.

Preparation

  • Equilibrate the column or resin with the provided endotoxin-free equilibration buffer to stabilize the ligand matrix.

Sample Loading

  • Our proprietary buffers help dissociate masked LPS-protein complexes, allowing the ligands to access and bind the Lipid A moiety.

Adsorption

  • The sample is processed via gravity flow, spin centrifugation, or FPLC. Endotoxins are sequestered within the polycationic network.

Collection

  • Elute target molecules and collect the fragment.

Regeneration

  • For bulk resins, use validated depyrogenation protocol to regenerate the column for multiple uses without ligand leakage.

FAQs

How do your kits handle "masked" endotoxins that standard LAL assays can't find?

Our kits include specialized dissociation buffers that break the hydrophobic and electrostatic bonds between LPS and your target protein.

Can I use these kits for acidic proteins that usually bind to anion exchangers?

Yes. Our multi-modal ligands provide high selectivity. By adjusting the pH close to the isoelectric point (pI) of your protein, we minimize competition and achieve high recovery rates.

How does the yield compare to ultrafiltration methods?

Ultrafiltration often results in significant loss if the protein is small or LPS monomers are present. Our kits consistently provide yields >90% across a wide range of sample types.

Are your kits compatible with high-salt buffers used in FPLC?

Our advanced adsorption chemistry is significantly less dependent on ionic strength than traditional methods, maintaining efficacy in up to 0.5 M NaCl.
