Recombinant Cascade Reagent Kits

Introduction Regulations Assay Principle Our Products Contact Us FAQs

Introduction

The Recombinant Cascade Reagent (rCR) represents the most advanced evolution in Bacterial Endotoxins Testing (BET). As global manufacturing scales, the limitations of natural resources, including biological variability and ecological impact, have necessitated a shift. The rCR assay is a sustainable, non-animal-derived alternative that utilizes three key cloned zymogens: Factor C, Factor B, and Proclotting enzyme. Unlike first-generation recombinant reagents that use only a single enzyme, rCR is engineered to simulate the full natural enzymatic cascade. Our Recombinant Cascade Reagent Kits help you achieve absolute endotoxin specificity and ensure long-term supply chain security through a biomimetic three-factor recombinant enzymatic pathway. Creative Biolabs utilizes advanced DNA technology to deliver an ethical, sustainable, and high-performance solution that mirrors the natural LAL cascade without the ecological footprint.

Regulation of the rCR Assay

The global regulatory landscape has evolved to support the adoption of recombinant technologies. Major pharmacopeias have established frameworks for using rCR as a suitable alternative to natural amebocyte lysate. Guidance under USP <85> and the implementation of USP <86> provide a clear path for using recombinant reagents in compendial testing. The European Pharmacopoeia has formally recognized the use of recombinant Factor C and cascade reagents, facilitating their use in final product release across Europe. Creative Biolabs provides all necessary documentation to assist your facility in meeting these international standards.

rCR Assay Principle

The principle of our rCR kit is a biomimetic reconstruction of the natural Limulus coagulation pathway. It functions through a kinetic chromogenic reaction:

Endotoxin Binding

  • When bacterial endotoxins enter the system, they bind to rFC, converting it to its active form.

Amplification

  • Activated rFC triggers rFB, which then activates the recombinant Proclotting enzyme. This multi-step process provides natural signal amplification.

Detection

  • The resulting active enzyme cleaves a specific chromogenic substrate, releasing yellow-colored p-nitroaniline (pNA).

Quantification

  • Endotoxin concentration is quantified photometrically by measuring the time to reach a specific optical density threshold.

Our Recombinant Cascade Reagent Kit Products

Creative Biolabs offers a range of Recombinant Cascade Assay Kits tailored to different laboratory scales and sensitivities. Unlike single step rFC assays that rely on fluorescence, our rCR kit leverages the natural enzymatic amplification of the cascade.

Superior Signal-to-Noise Ratio

The multi-step process naturally amplifies the signal, allowing for high sensitivity (<0.005 EU/mL) using standard absorbance readers at 405 nm.

Enhanced Environmental Recovery

Cascade-based reagents achieve significantly higher recovery rates for environmental endotoxins compared to single-factor rFC method.

Reduced Interference

The presence of Factor B and the full cascade architecture provide a more robust reaction environment, often allowing for lower Non-Interfering Dilutions (NID) in complex matrices like high-protein or high-salt samples.

FAQs

Is the rCR assay as sensitive as traditional LAL?

Yes. Our kits offer sensitivity down to 0.001 EU/mL, matching or exceeding the performance of the most sensitive LAL reagents currently available.

Do I need to buy new equipment to use this kit?

No. Our Recombinant Cascade Reagent Kit is designed for standard kinetic chromogenic platforms. If you have an incubating microplate reader capable of measuring at 405 nm, you are ready to start.

How does rCR eliminate false positives?

False positives in LAL are typically caused by the Factor G pathway reacting with glucans. Since rCR is synthetically engineered without Factor G, this entire interference pathway is removed.

Can I use this for environmental water monitoring?

Our rCR technology is highly effective at detecting autochthonous (wild) endotoxins in water systems with recovery rates comparable to LAL.
