Limulus Amebocyte Lysate (LAL) Reagent Water Products

Introduction Standards Applications Our Products FAQs

Introduction

In the stringent landscape of pharmaceutical manufacturing, the bacterial endotoxins test (BET) serves as a critical safety checkpoint. The success of this enzymatic assay depends entirely on the solvent used to reconstitute reagents and dilute samples. LAL reagent water (LRW) is specially processed, pyrogen-free water specifically validated to show no detectable endotoxin and no interference with the LAL enzyme cascade. While often confused with standard Water for Injection (WFI), the LRW is held to a much more stringent standard to ensure it contains neither endotoxins nor (1,3)-β-D-glucans.

Global Standards

According to USP <85>, LRW is defined as water that shows no reaction with the specific LAL reagent with which it is to be used.

Specification LAL Reagent Water (LRW)
Endotoxin Concentration < 0.001 EU/mL or < 0.005 EU/mL (Depending on sensitivity)
Glucan Content < 1.56 pg/mL (Glucan-free)
pH Range 6.0 – 8.0 (Optimized for LAL Enzyme Activity)
Packaging Validated depyrogenated glass or plastic

Applications of LAL Reagent Water

LRW is an indispensable reagent that facilitates every stage of the endotoxin detection protocol:

Reagent Reconstitution

It is used to restore lyophilized LAL lysate and control standard endotoxin (CSE) to their functional liquid states.

Standard Curve Preparation

It is critical for creating serial dilutions of endotoxin standards. Any baseline contamination in the water would skew the linearity of the curve, leading to inaccurate quantification.

Sample Dilution and Extraction

LRW is used to dilute samples to the maximum validated dilution (MVD) in complex drug matrices testing or to extract pyrogens from device surfaces and lumens.

Our LAL Reagent Water Products

Creative Biolabs provides the high-performance LAL reagent water necessary for the most demanding pharmaceutical and medical device applications. By combining ultra-low endotoxin thresholds with glucan-free certification and professional technical support, we empower your lab to achieve absolute data integrity.

Ultra-Low Endotoxin Levels

Maintaining a baseline significantly below the assay's limit of detection.

Glucan-Free Status

Ensuring no cross-reactivity with the glucan-sensitive factor G protein.

Ionic Stability

Precise control over divalent cations and heavy metals that can inhibit or unnaturally enhance the enzymatic reaction.

Optimal pH Range

Maintaining a stable pH between 6.0 and 8.0, which is the sweet spot for LAL enzyme activity.

FAQs

Can I use standard WFI instead of LAL Reagent Water?

While WFI is pure, it is only required to be < 0.25 EU/mL. If your assay sensitivity is 0.03 EU/mL or lower, standard WFI may interfere with your results. Our LRW is specifically validated to < 0.005 EU/mL to eliminate this risk.

How does glucan contamination affect my LAL test?

Glucans can trigger the factor G pathway, which mimics an endotoxin reaction and leads to false positives. Creative Biolabs' LRW is certified glucan-free, ensuring your test is 100% specific to bacterial endotoxins.

What is the shelf life of LAL Reagent Water once opened?

To maintain the highest purity, we recommend using LRW immediately upon opening. If stored, it should be tightly sealed and kept at 4°C, though we suggest single-use vials for the most sensitive kinetic assays to prevent ambient contamination.
