The Recombinant Cascade Reagent (rCR) is an animal-free method used for the quantitative detection of gram-negative bacterial endotoxins (lipopolysaccharides, or LPS). rCR fully mimics the natural, multi-step enzymatic cascade found in the blue blood of the horseshoe crab (Limulus polyphemus), using three genetically engineered recombinant proteins that act sequentially to amplify the detection signal. Step 1 Activation of Recombinant Factor C: The cascade begins when endotoxin molecules in a sample bind to inactive recombinant Factor C (rFC). This binding causes a conformational change that activates rFC into an active protease. Step 2 Activation of Recombinant Factor B: The activated recombinant Factor C then targets inactive recombinant Factor B (rFB). It proteolytically cleaves rFB, converting it into its enzymatically active form. Step 3 Activation of Recombinant Proclotting Enzyme: The activated recombinant Factor B in turn recognizes and cleaves the inactive recombinant proclotting enzyme, transforming it into the active recombinant clotting enzyme. Our Recombinant Cascade Reagent Endotoxin Detection Kit (ES-0526-YC200) was designed to seamlessly replace traditional LAL. This kit offers an eco-sustainable, highly precise, and animal-free solution for your quantitative endotoxin testing needs.
