The Principle of Kinetic Turbidimetric Detection
When bacterial endotoxin interacts with the amebocyte lysate reagent, it initiates an enzymatic cascade that leads to the cleavage of coagulogen, forming an opaque gel clot. A microplate reader continuously monitors the optical density (OD) of the mixture at a specific wavelength (340 nm or 405 nm). The time required for a sample to achieve a specific, predefined increase in absorbance is negatively correlated with the endotoxin concentration.
