The biochemical foundation of the Kinetic Turbidimetric Kit relies on the initial stages of the natural Limulus coagulation cascade. When an endotoxin is introduced into the mixture, it triggers a multi-stage proenzyme cascade that culminates in the activation of the core Clotting Enzyme. This enzyme cleaves the natural protein coagulogen inherent in the lysate, altering its structural solubility. As a result, the solution becomes progressively turbid as it moves toward a physical gel state. An incubating microplate reader continuously monitors the light transmittance of each well at 405 nm or 340 nm at regular intervals throughout the incubation process. The incubation software measures the time required for a specific well's optical density (OD) to rise to a pre-set absorbance threshold.
