Separation Principle:
A combination of biotin-conjugated antibodies and anti-biotin microbeads is employed for the indirect magnetic labeling of non-CD4⁺ T cells. Thus, a high level of purity in untouched T cells can be isolated by depleting the magnetically labeled non-CD4⁺ T cells.
Background Information:
The CD4 antigen is expressed on most thymocytes and mature T helper cells, and at lower levels on a subpopulation of dendritic cells. CD4 acts as an accessory molecule in the recognition of MHC class II/peptide complexes by the TCR heterodimer on CD4⁺ T helper cells.
