This product is an antibody that was generated by immunizing rabbit against a peptide located in the N-Terminal extracellular domain of Frizzled 5 (Human). It is used only in in vitro assays.

Introduction: The Frizzled 5 protein is believed to be the receptor for the Wnt5A ligand. Frizzled 5 has been reported to be expressed in fetal kidney, fetal and adult liver, fetal lung, and adult pancreas. ESTs have been isolated from bone, liver/spleen, placenta, and prostate libraries. Frizzled 5 was cloned from a retina cDNA library. Receptor for Wnt proteins. Component of the Wnt-Fzd-LRP5-LRP6 complex that triggers beta-catenin signaling through inducing aggregation of receptor-ligand complexes into ribosome-sized signalosomes. The beta-catenin canonical signaling pathway leads to the activation of disheveled proteins, inhibition of GSK-3 kinase, nuclear accumulation of beta-catenin and activation of Wnt target genes.

  1. Q 1: How do I ensure the specificity of the Frizzled 5 polyclonal antibody in my experiments?

    A: With gentle agitation at primary antibody incubation and appropriate dilution as well as diluent, specificity will be achieved when using the provided polyclonal Frizzled 5 antibody. The bands of the predicted size for Frizzled 5 should be observed around 65 kDa in the applications of Western blot. The identity of such additional bands, for example, those apparent at 48 kDa, can be ambiguous and should be confirmed with appropriate control peptides or competition experiments of the peptide to assess specificity.

  2. Q 2: Can I use the Frizzled 5 polyclonal antibody for applications beyond Western blotting?

    A: Yes, the polyclonal antibody against Frizzled 5 is versatile, applicable in many fields like Immunofluorescence (IF), Immunohistochemistry (IHC), and ELISA, done by them. Validations have been done with human, mouse, and rat species. Ideally, one would prefer retrieval of antigens in Tris-EDTA buffer (pH 9.0), which is shown to improve staining quality. Adding to that, one should choose proper sources and detection systems for their secondary antibodies.

  • Reliable Detection in Cellular Models
    Frizzled 5 polyclonal from Creative Biolabs was quite an instrumental ab against Frizzled 5, which we characterized in the project as we were working out the signaling pathways unmasked in HepG2 cells. And since the antibody was very efficient in its job with few ambiguities in the case of immunofluorescence tests, working with PBMC cells only helped us to gain some extent of consistency with clear labeling of Frizzled 5, sometimes reinforcing the clarity of what was observed. This, in fact, will not only increase the confidence and hence reliability of the generated experimental findings but, more importantly, shine light on the utility of the antibody for complex cellular studies.
