The Enzyme Cascade The biochemical foundation of the Creative Biolabs Gel Clot LAL Assay relies on a highly sensitive, proenzyme amplification cascade naturally present within the amebocyte cells of the Atlantic horseshoe crab. When the sample is incubated at 37°C, the activated Clotting Enzyme cleaves the specific protein coagulogen inherent in the lysate. This alters its structural solubility, giving rise to an opaque gel clot. A definitive positive result is confirmed if the gel remains firmly intact at the bottom of the tube after a smooth, single-motion 180° inversion.
