Introduction of SLITRK6

SLITRK6 is an integral membrane protein known to have high expression levels in certain carcinomas. The gene has an open reading frame of 841 amino acids (aa) and was identified as SLITRK6, a member of the SLITRK family of proteins. The SLITRK family members are type I transmembrane proteins that share conserved leucine-rich repeat domains similar to those in the secreted axonal guidance molecule, SLIT. They also show similarities to Ntrk neurotrophin receptors in their carboxy-termini, sharing a conserved tyrosine residue.This protein was originally discovered as a bladder tumor antigen using suppressive subtractive hybridization (SSH) on biopsies from bladder cancer patients. Extensive immunohistochemistry showed that SLITRK6 is expressed in multiple epithelial tumors, including bladder, lung, and breast cancer as well as in glioblastoma. Examination of 509 formalin fixed paraffin embedded (FFPE) biopsies, representing all stages of bladder cancer showed that SLITRK6 is expressed in 90% of urothelial carcinomas, 100% of metastatic bladder cancers.

Anti-SLITRK6 ADC in Urothelial Tumor

Studies have shown that SLITRK6 is highly expressed in bladder cancer and other tumors of epithelial origin. Scientists have developed ASG-15ME, a human monoclonal antibody (mAb) conjugated to MMAE through a protease-cleavable linker, to target SLITRK6. ASG-15ME induced potent inhibition, and even tumor regression, in multiple xenografts. Findings reveal that ASG-15ME shows excellent antitumor activity in vivo with significantly better activity. ASG-15ME is currently in phase I clinical trials for the treatment of metastatic urothelial cancer.
