CD72

CD72 molecule (CD72), also known as LYB2, CD72b, is a 40‑45 kDa type II transmembrane glycoprotein expressed mostly in B cells. Mature human CD72 contains a 95 amino acid (aa) cytoplasmic domain with two immunoreceptor tyrosine‑based inhibitory motifs (ITIMs), a 21 aa transmembrane segment, and a 243 aa C-type lectin-like domain (CTLD) in the extracellular region. It is report that the ECD of human CD72 shares 48% and 44% amino acid sequence identity with that of mouse and rat CD72, respectively. Besides expressed on B lineage cells, this protein is also expressed in NK cells, monocytes, dendritic cells and mast cells.

By recruiting SH2 domain–containing phosphatase 1 (SHP-1) to the phosphorylated ITIM, CD72 negatively regulates B cell antigen receptor (BCR) signaling. In addition to its negative regulation of BCR signaling, CD72 can bind to Semaphorin 4D and induce cytokine production by monocytes and dendritic cells, inhibits SCF R/c‑kit induced mast cell proliferation and activation, and inhibits the cytolytic activity of NK cells
