FLT1

FLT1, also named as VEGFR1, is an elusive member of the VEGFR family. The VEGFR family contains three receptors: FLT1, FLK1 and FLT4 (also known as VEGFR1, VEGFR2 and VEGFR3, respectively). Among them, FLK1 binds VEGFA and VEGFC, and it is the major receptor which drives angiogenesis in healthy conditions and during disease. FLT4 binds VEGFC and VEGFD. However, FLT1 binds VEGFA, VEGFB and PlGF. FLT1 and FLK1 comprise seven extracellular immunoglobulin (Ig)-like domains, an intracellular tyrosine-kinase domain, and a trans-membrane region.
In general, FLT1 is upregulated in ischaemic and inflammatory diseases, and in a variety of tumours, such as prostate, non-small cell lung cancer (NSCLC), pulmonary adenocarcinoma, nephroblastoma, breast and colon cancer, glioblastoma, hepatocellular carcinoma, and multiple myeloma. Moreover, FLT1 is also expressed via a number of malignant cells, for example, breast and leukaemic cancer cells, oesophageal, and prostate. It has shown that FLT1 immunoreactivity in tumour cells indicates poor prognosis, metastasis, and recurrence of NSCLC and breast cancer. Unlike breast cancer and NSCLC, leukaemic cells that express FLT1 have a more invasive and aggressive phenotype in vivo.
