IL13RA2

Interleukin 13 receptor subunit alpha 2 (IL13RA2), also known as CT19, IL-13R, IL13BP, or CD213A2 (cluster of differentiation 213A2), is a membrane protein that exerts a high expression level in testis in healthy humans. It shows sequence similarities to the type I cytokine receptor family and it belongs to the subfamily type 5. IL13RA2 contains a WSXWS motif, which has proven to be important for correct protein folding and subsequent intracellular transport and cell-surface receptor binding. Meanwhile the "box 1" motif in IL13RA2 is crucial for JAK interaction and/or activation.

IL13RA2 is closely related to IL13RA1, another subunit of the interleukin 13 receptor complex. Monomeric IL13RA2 exerts high affinity interaction with interleukin 13 (IL13) and is reported to mediate IL13internalization. In the meantime, it lacks the cytoplasmic domain and does not serve as signal mediator of IL13. IL13RA2 is a potential target for breast cancer and due to its capacity for internalization, it is a promising target for ADC-based immunotherapies.
