IL17A also known as IL17 or CTLA8, is a protein that in humans encoded by the IL17A gen. IL17 is a proinflammatory cytokine formed by activated T cells. The initiate sources of IL-17 are lymphocytes, such as gamma-delta T (γδ-T), CD4+, CD8+, innate lymphoid cells (ILCs) and invariant NKT. Besides, Non-T cells have also been found to generate IL-17 in certain situations. IL17 adjusts the activities of NF-kappaB and mitogen-activated protein kinases. This cytokine also enables to improve the expression of IL6 and cyclooxygenase-2 (PTGS2/COX-2), and increases the generation of nitric oxide (NO).
IL-17A is reported to be associated with host defense against a variety of pathogens. The inappropriate generation of IL-17A is regarded to be related to the progress of Th17-mediated autoimmune and inflammatory conditions, including rheumatoid arthritis, multiple sclerosis, as well as psoriasis. Importantly, IL-17 serves an essential role in inflammation, autoimmune diseases and host defense, which results in the experimental targeting of the IL-17A pathway in animal models of diseases and in clinical trials in humans.
