IL4R

IL4R also called CD124, is a type I transmembrane protein that enables to bind interleukin 4 (IL4) and interleukin 13 to modulate IgE production. IL4R also can bind interleukin 4 to enhance the differentiation of Th2 cells. IL4R was initially found in murine sarcoma and colonic adenocarcinoma cells. Human gliomas have also been found to produce high levels of IL-4R mRNA and protein. Besides, a soluble form of IL4R is able to be formed via proteolysis of the membrane-bound protein, and this soluble form is capable of inhibiting IL4-mediated cell proliferation and IL5 upregulation by T-cells.
It has also reported that the N-terminal (extracellular) portion of IL4R is associated with overall topology to fibronectin type III modules and folds into a sandwich containing seven antiparallel beta sheets arranged in a three-strand and a four-strand beta-pleated sheet. They play an essential role in the binding of IL4 to the receptor alpha chain, which is an important event for the formation of a Th2-dominated early immune response. In addition, allelic variations in IL4R gene have been related to atopy, a condition which enables to manifest itself as allergic rhinitis, sinusitis, asthma, or eczema.
