Integrin subunit alpha L (ITGAL), also known as lymphocyte function-associated antigen-1 (LFA-1), encoded by ITGAL gene, belongs to the integrin family which consists of an alpha chain and a beta chain. It is widely expressed on the surface of T lymphocytes, B lymphocytes, monocytes, macrophages and neutrophils. Previous studies have been shown that LFA-1 interacts with its ligands, molecules 1 through 3 (ICAMs 1-3), regulating leukocyte intercellular adhesion. In addition, the coordinated stimulus signal is released via the combination of LFA-1 and ICAM-1 to promote lymphocyte activation, proliferation and differentiation. These suggest that LFA-1 plays an important role in cell intercellular adhesion and immune responses.
The LFA-1 is selectively expressed in multiple tissues, including appendix, bone marrow, lymph node, spleen, gallbladder, small intestine and so on. It has been suggested that the changes of LFA-1 expression may be associated with the processes of various diseases, such as rheumatoid arthritis, acute megakaryoblastic leukemia, transient myeloproliferative, multifocal leukoencephalopathy, immune thrombocytopenia. For example, the deficiency of LFA-1 is an indicator of acute megakaryoblastic leukemia and transient myeloproliferative disease.
