LYPD3

LY6/PLAUR domain containing 3 (LYPD3) is often referred to as C4.4A. It is comprised of ~350 amino acids and with a molecular weight at 36 kDa. The protein contains two UPAR/Ly6 domains and it is modified with both N- and O-glycans via post translational modifications. Normal LYPD3 expression is observed in placenta, skin, and urothelium. It is shown to bind laminin-1 and laminin-5. Other molecules that interact with LYPD3 include LGALS3, AGR2, and AGR3.

LYPD3 plays a role in cell migration and urothelial cell-matrix interactions. Abnormalities of LYPD3 are observed in primary and metastatic transitional cell carcinomas (TCCs), in breast cancer, as well as in the tumor component of some noninvasive superficial lesions. High expression of LYPD3 has also been reported in invasive and metastatic urothelial cancers. LYPD3 is proposed to involve in promoting tumor progression and it serves as a good target for immunotherapies.
