Syndecan 1 (SDC1), also known as CD138, SDC, SYND1 or syndecan, is a glycosylated, type I transmembrane protein and it is a member of syndecan proteoglycan family. The precursor of Syndecan 1 is 310 amino acid (aa) in length, containing a 17-aa signal sequence, a 234-aa extracellular domain (ECD), a 25-aa transmembrane segment, and a 34-aa cytoplasmic region, which is further processed into a mature and soluble form by cleaving the membrane-bound ECD.

With functions as an integral membrane protein, the syndecan-1 protein participates in cell proliferation, cell migration and cell-matrix interactions via its receptor for extracellular matrix proteins. Altered syndecan-1 expression has been reported in several different tumor types. Moreover, this protein is highly overexpressed on multiple solid tumors and hematological malignancies, and represents one of the most specific target antigens for identification of multiple myeloma (MM) cell. Notably, it is the target of Indatuximab ravtansine, an antibody-drug conjugate (ADC) in clinical study consisting of the anti-CD138 chimerized MAb (nBT062) linked to DM4.
