Transmembrane protein with EGF like and two follistatin like domains 2 (TMEFF2), also known as TR, HPP1, TPEF, TR-2, TENB2, or CT120.2, is a tomoregulin family protein that is localized at cell membrane. It contains one EGF-like domain and two Kazal-like domains in the protein sequence and it is post translationally modified by N-glycosylation. TMEFF2 undergoes proteolysis (proteolytic shedding) that produces a soluble form, namely TMEFF2-ECD.
TMEFF2 is actively expressed in brain, spinal cord and prostate. It functions either as an oncogene or a tumor suppressor depending on the stimuli and it is demonstrated to mediate prostate cancer cell invasion. The shedded form, TMEFF2-ECD, is carcinogenic since it up-regulates cancer cell proliferation via the promotion of ERK1/2 phosphorylation. TMEFF2 is highly expressed in prostate carcinoma, making it a target against this cancer via different forms of immunotherapy.
