The complement system is a part of the immune system that triggers immune functions as phagocytosis, inflammation and membrane attack, in which complement component 3 (C3) is the convergent point in complement pathways. Detection of C3 autoantibody (anti-C3), a kind of natural autoantibody (NAA), may be associated with some autoimmune diseases. Background of Anti-C3

C3 is the most important and abundant protein in the complement system, which plays a significant role in immune responses, immune complexes clearance, and cell disruption. C3 is the convergent point of the complement system pathways including the classical, lectin, and alternative pathways. C3 and the receptor of C3b on erythrocytes are important proteins that process and transport large immune complexes from the circulation. Due to the importance, autoantibodies react to C3 (anti-C3) may relate to pathological consequences. Determination of both C3 and anti-C3 levels, combining comparison with other complement components and normal individuals are helpful in diagnosis and monitoring treatment of certain diseases.

The Role of Anti-C3 in Systemic Lupus Erythematosus

Autoimmune diseases are mostly caused by autoantibodies induced by an inefficient clearance of apoptotic cells and cellular debris, which abnormally serve as a source of autoantigens attacking healthy tissues in many parts of body-self. Systemic lupus erythematosus (SLE) is an autoimmune multisystem disease which is characterized by multiple autoantibodies and complement activation. Various autoantibodies have been found in SLE typically as anti-nuclear antibody, anti-double-stranded DNA, anti-rheumatoid factor, etc. Antibodies against C3 (Anti-C3) have been described in animal models and SLE patients by many investigations. Researches propose that anti-C3 may be one of the biomarkers and immunological hallmarks of SLE, and it is helpful in the early and rapid diagnosis.
