Thioredoxin Library Construction

Creative Biolabs proudly offer the unique thioredoxin (TrxA) library construction service for our customers all over the world. Through our proprietary HiAffi™ platform, high quality thioredoxin library can be generated with higher diversity.

Thioredoxin (TrxA) is a kind of oxidoreductase containing a dithiol-disulfide active site. It is ubiquitous and known to be present in almost all organisms. In Escherichia coli, thioredoxin is an enzyme involved in maintaining the cytosolic thiol/disulfide equilibrium inside the cell. The properties are recognized as small size (12 kDa), highly soluble, structurally robust and can be overexpressed in large amounts. In addition, the short active site of thioredoxin forms a solvent-accessible loop in the oxidized state which permits the insertion of diverse and quite long peptide stretches. This represents a unique fashion of providing structural randomness.

By replacing its active site loop with an inserted random peptide sequence, thioredoxin can be employed as a scaffold for the display of single conformationally constrained peptides. Moreover, for peptides which may not be singly useful due to the small size or other structural limitations, the development of thioredoxin scaffold offers a reliable solution for their potential application. Combining with appropriate display technology, thioredoxin scaffold libraries are able to be generated to achieve extensive diversity, which the thioredoxin variants (dubbed peptide aptamers) could be selected for different targets and useful in various applications.

To achieve high quality scaffold library, Creative Biolabs has developed the unique Hi-Affi™ phage display platform for our library generation services. This platform is improved from phage display technology, while has combines trimer codon technology and NNK method for better diversity and affinity. In general, our scientists can obtain thioredoxin libraries with 100% precise mutant and over 1010 diversity.
