Background of AntI-Na+/I- Symporter Antibody

Iodide (I-) is a scarce environmental microelement that is a vital component of thyroid hormone. Cellular uptake of I- is mediated by NIS, a plasma membrane glycoprotein which transports two Na+ ions per each I- ion and plays a key role in the functioning of the thyroid gland. NIS belongs to the sodium-dependent transporter family 5A. NIS is an integral membrane protein with the amino-terminus facing the extracellular milieu and carboxyl-terminus facing the cytoplasm. The presence of IgG with NIS reactivity suggests that these autoantibodies might affect thyroid function by inhibiting the uptake of I-, thus playing a role in hypothyroidism.

The Role of AntI-Na+/I- Symporter Antibody in Thyroiditis

NIS antibodies are present in a proportion of patients with AITD and thus represents an important autoantigen in AITD. Scientists reported that autoantibody against NIS was found in the sera of 84% of patients with Graves' disease (GD) and 12% of patients with Hashimoto's thyroiditis (HT). It is suggested that the incidence of the autoantibodies against NIS is higher in GD than HT, and these antibodies may inhibit I- uptake.
