Background of Anti-thyroglobulin Antibody

Tg is a vertebrate secretory protein synthesized in the thyrocyte endoplasmic reticulum (ER). A combination of frameshift mutations, nonsense mutations, missense mutations, and splice site mutations in Tg occurs spontaneously to cause congenital hypothyroidism and thyroidal ER stress. Anti-Tg antibody (TgAb) is induced due to coexistent lymphocytic thyroiditis (LT) or the papillary thyroid carcinoma (PTC) per se and are present in the serum of approximately 20% of patients with differentiated thyroid diseases. The prevalence of TgAb is significantly higher in PTC than in follicular thyroid cancer. TgAb can develop in patients with subacute thyroiditis (SAT). Now, serum TgAb, the marker of several kinds of thyroiditis and residual tumor in PTC, can be estimated to promote the disease diagnosis.

The Role of Anti-thyroglobulin Antibody in Thyroiditis

Hashimoto's thyroiditis (HT), an autoimmune thyroid disease, is one of the most widespread thyroid disorders. HT is characterized by the presence of elevated circulating antibodies, especially anti-thyroid peroxidase (anti-TPO) and anti-Tg. High titers of TgAb have been found in approximately 90% of HT patients, hence TgAb becomes a diagnostic serological marker of HT. In vitro experiments have shown that TgAb had an effect on antibody-dependent cellular cytotoxicity (ADCC), which indicated that it might be involved in thyrocyte destruction. Therefore, TgAb IgG represents a hallmark of this debilitating autoimmune disease.
