Fagellin FliC
Flagellin is the primary component of bacterial flagella. The immunogenicity of bacterial flagellin has been reported in numerous studies. By its close interaction with the immune system, the flagellin represents an interesting adjuvant and vaccine candidate. Flagellin of Salmonella induces several pathways of immunity system, which leads to producing antibody and cytokines. As for our target, FliC could stimulate both cellular and humoral immune responses. The fliC and fljB genes encoding the two flagellar antigens present in Salmonella. Salmonella can undergo flagellar phase variation to alternately express these two different major flagellar antigens, phase 1 and phase 2, encoded by the fliC and fljB genes, respectively.

Structure of FliC
Flagella FliC is a virulence factor and protective antigen. The folded Salmonella flagellin protein assumes an “L” shaped structure, comprised of four unique domains, D0, D1, D2, D3. The 250 amino acids have been documented as mostly invariant among Gram-negative and Gram-positive bacteria. The unique protein epitopes present within Salmonella flagellins are characteristic and conserved for individual serovars and provide the basis in part for serotyping in the Kauffman-White scheme.

Antibody of FliC
Studies have shown a high prevalence of Salmonella in various animals and humans. These strains usually possess high levels of drug-resistance. Monoclonal antibodies (mAbs) against flagellin have been shown to mediate protection in animal models against infections caused by Salmonella. The immunostimulatory properties of flagellin have further improved immune responses towards co-administered protein antigens after admixture, covalent linkage, or incorporation into non-covalent complexes. So, combining an approach designed to enhance the host immune response with antibiotic therapy against the drug-resistance pathogen is a promising therapy.
