Background of Anti-Aβ Antibodies

It is known that the amyloid-beta (Aβ) peptide is a 40/42 hydrophobic amino acid peptide cleaved from a transmembrane amyloid precursor protein (APP) that tends to polymerize. Although the specific bio-functions still remain not so clear, it has been revealed that Aβ seems to be closely involved in Alzheimer's disease (AD). Several assemblies of Aβ are detected in the brain of AD patients, including soluble Aβ monomers, oligomers and insoluble Aβ fibrils in amyloid plaques and amyloid vascular deposits. The Aβ oligomers appear to be the most neurotoxic in the three assemblies, causing damage and death of neurons in the brain of patients with AD. Previous reports suggested that passive immunization of AD patients using intravenous immunoglobulin (IVIg), which contains natural anti-Aβ antibodies, improves cognitive function in AD patients. These natural anti-Aβ antibodies contribute to preventing Aβ-induced neurotoxicity. This neuroprotective effect may lay the foundation of diagnostic value and the therapeutic potential of the anti-Aβ antibodies for the treatment of AD.

The Role of Anti-Aβ Antibodies in Alzheimer's Disease

Alzheimer's disease (AD), a type of dementia, is a chronic neurodegenerative disorder that usually begins with hypomnesia and worsens over time leading to severe memory impairment even other mental disabilities. AD presents mental deterioration with diffuse neuronal loss and is pathologically identified by Aβdeposition and neurofibrillary tangles in the brain. Autoantibodies specific against Aβ could be detected in the cerebrospinal fluid and plasma of both healthy individuals and AD patients, however, the levels of the unbound Aβ autoantibodies in AD patients were lower than that in healthy controls. Meanwhile, it was demonstrated that the levels of antigen-bound Aβ-autoantibodies in AD patients, namely Aβ-IgG immune complexes, were remarkably higher than in controls, which exhibited a negative correlation with the progression of AD. Specifically, these revealed anti-Aβ autoantibodies mainly are human IgM- and IgG-specific antibodies, and the specific activity of IgM anti-Aβ antibodies in human serum averaged 2.5-fold higher than the specific activity of IgG anti-Aβ antibodies. Therefore, Aβ autoantibodies, including anti-Aβ IgM and anti-Aβ IgG, can serve as blood-derived biomarkers applied for AD clinical diagnosis.
