Background of Anti-HSP 70 Antibodies

Heat shock protein 70s (Hsp70s) are a kind of molecular chaperones that highly conserved and ubiquitous in most of the organisms, which play important roles in cellular stress and various cellular processes, including protein folding, protein translocation, and protein degradation. It was reported that antibodies to Hsp70 have been isolated from dilated cardiomyopathy (DCM) patients. Other studies found that antibodies against HSP 60 and 70 at higher frequency in DCM sera than in ischemic or normal control subjects. These results suggested that a complement-dependent, antibody-mediated mechanism of damage to cardiac myocytes may contribute to the pathogenesis of myocarditis/DCM.

The Role of Anti-HSP 70 Antibodies in Myocarditis

Myocarditis is an inflammatory disease occurring in heart muscle, which can cause acute heart failure, dilated cardiomyopathy, even sudden death. Evidence demonstrated that autoantibodies against HSP 70 were significantly increased in the sera of patients with myocarditis or DCM. Besides, these anti-HSP 70 antibodies were found mainly existing in young patients compared with older people. Therefore, these antibodies are of pathogenetic significance or may be used as a disease marker to promote the diagnosis related to cardiovascular disease.

The Role of Anti-HSP 70 Antibodies in Nasopharyngeal Carcinoma

Nasopharyngeal carcinoma (NPC) is the most frequent cancer arising in the nasopharynx, which is one of the five main types of head and neck malignant tumor. During the NPC processes, Hsp 70 has been found involved in the cancer progress with different expression patterns. Furthermore, autoantibodies against HSP70 also have been produced in NPC patients. In addition, the concentrations of HSP70 and anti-HSP70 antibodies in nasal secretions (NS) have also been evaluated in patients with chronic rhinosinusitis (CRS), and results showed that the NS levels of HSP70 and anti-HSP70 antibodies were increased in subjects with CRS compared with healthy controls. These results suggested that Hsp 70 and anti-HSP70 antibodies could be useful biomarkers in the diagnosis of NPC and CRS.

The Role of Anti-HSP 70 Antibodies in Gastric Cancer

Gastric cancer, also referred to stomach cancer, is a disease in which malignant cells form in the lining of the stomach. HSP70 significant over-expression has been observed in early gastric cancers. Increased HSP, acting as tumor-related antigens, further stimulated the immune system to produce HSP autoantibodies. The anti-HSP 70 autoantibody were detected in sera of patients with gastric cancer, and the concentrations of serum Hsp70 autoantibody were significantly higher for patients with gastric cancer (mean, 0.236 ± 0.112 mg/mL, P < 0.001) than healthy individuals (0.207 ± 0.055 mg/mL, P < 0.001). These sera biomarkers analysis gave us a hint that Hsp70 autoantibody might be served as one of the markers for the gastric cancer diagnosis.
