Background of Anti-laminin Antibody

Laminin is a group of large heterotrimeric proteins composed of three types of peptide chain: α, β, and γ with different structures and functions. Laminin is the first extracellular matrix glycoproteins detectable in the embryo, which is also a major component of the basement membrane. In structure, cross-shaped laminins are the basis of protein networks in most cells and organs, which lays the foundation for the importance of the basal lamina in influencing cell differentiation, migration, and adhesion.

Various combinations of chains form different laminins and at least 16 distinct laminin isoforms have been identified. Anti-laminins antibodies are a group of corresponding autoantibodies targeting to laminins. Anti-laminins antibodies and abnormality of these laminins are associated with a wide range of human diseases including cancer, infections, heart diseases, and autoimmune disorders.

The Role of Anti-laminin in Myocarditis

Myocarditis is a kind of inflammation of the heart muscle (myocardium), which may cause degeneration or death of heart muscle cells. Myocarditis may be caused by many disorders (e.g. viral infections, bacterial infections, autoimmune disorders, cardiotoxins, drugs) but is often idiopathic. Symptoms can vary and can include fatigue, dyspnea, edema, abnormal heartbeat, breath, chest pain, and sudden death. Diagnosis is often based on symptoms and clinical findings of abnormal ECG, cardiac biomarkers, and cardiac imaging in the absence of cardiovascular risk factors.

It has been widely reported that several circulating autoantibodies induced by infections play a crucial role in the progression and induction of myocarditis. In addition to anti-sarcolemmal antibody and anti-myolemmal antibody, increased circulating anti-laminin antibodies can be observed both in incidence and concentration of myocarditis, perimyocarditis and dilated cardiomyopathy. These high prevalences of anti-laminin antibodies raise the possibility of the diagnostic value to myocarditis.
