Overview

The most basic function of an antibody is to neutralize the target antigen, and other functions include cross-linking to induce soluble antigen precipitation or cell agglutination. Antibodies are one of the most commonly used tools in basic scientific research, in vitro diagnosis, and clinical testing. They were routinely used for Western blotting (WB), quantitative immunofluorescence (QIF), enzyme-linked immunosorbent assay (ELISA), immunoprecipitation (IP), and immunohistochemistry (IHC). They are also important tools for clinical management, widely used in laboratory medicine (ELISA assay and flow cytometry) and anatomical pathology (IHC). The significant impact of antibody therapy in disease treatment has led to the emergence of many new forms of macromolecules. Antibodies and antibody-derived macromolecules have become the main pillars of protein-based therapeutic molecules (biologics), which are widely used in the treatment of various diseases and disease states, including cardiovascular diseases, autoimmune diseases, malignant tumors, and infections.

Applications

The applications of high-throughput antibody expression and purification fall into two broad classes. For microscale applications, HT antibody purification provides a front-end for antibody production to meet various applications, such as analysis of protein interactions, enzyme substrate networks, etc., with limited antibody quality required for each sample. For applications that require a large amount of purified antibody (mg), the HT method for protein production can effectively screen for many different constructs to identify those candidates that have high activity and better characters. In these applications, success often depends on the time-consuming trial-and-error process of attempting to express different versions of the target protein until a well-expressing, soluble, and correctly folded construct can be identified. The ability to screen multiple structures simultaneously can greatly speed up the screening process.
