Introduction to Anti-Endothelial Cells (AECA) Antibody

Anti-endothelial cell antibodies (AECAs), are a group of autoantibodies that can bind to endothelial cells (ECs) by several portions of the immunoglobulins. AECAs with a wide variety of immunoglobulin isotypes, such as IgA, IgG, and IgM, have been reported, and the data suggests that these AECAs play an important role in mediating various inflammatory diseases, including systemic vasculitis, rheumatoid arthritis (RA) and systemic lupus erythematosus (SLE). In particular, AECA can regulate the EC activation, the induction of antibody-dependence cytotoxicity, as well as cell apoptosis in animal models. Meanwhile, recent researchers have demonstrated that AECA can enhance the interaction between polymorphonuclear leukocytes (PMNs) and ECs by activating FcγRIIIb and costimulatory molecules, for instance, CXCR-1 and CXCR-2. Additionally, many studies have been focused on identifying novel EC target antigens for AECA, which is essential to improve the ability of AECA detection and to understand AECA mechanisms of action. In this condition, many organizations or companies are dedicated to developing safe and effective AECAs by detecting antigen specificity, binding affinity. Additionally, a wide collection of AECA detection assays have been also developed in both in vivo and in vitro.

The Service of Anti-Endothelial Cells (AECA) Antibody

The pathogenic role of AECAs has been confirmed in a number of diseases, especially in many types of systemic vasculitis diseases. As a result, Creative Biolabs offers a full range of AECA services to help our client develop novel AECAs at cost-effective manners. We have established a panel of platforms to discovery potential antigens for AECAs. In general, a series of autoantigens derived from different subtypes of ECs, such as macrovascular ECs, lineage ECs, and microvascular ECs, have been generated for AECA development. Moreover, A number of assays have been used to detect AECA in various disease models, including enzyme-linked immunosorbent assay (ELISA), western blotting (WB), flow cytometry, fluorescence-activated cell sorting (FACS), and immunofluorescence (IF). In a recent study, an AECA has been detected in a mouse model of SLE by using a cell-surface ELISA on living cells. Microvessels and endothelial lineage cells are common cell substrates in our detection assays. In addition, the efficacy of these assays has been compared by our labs, and the results indicate that cellular ELISA assay should be more effective in identifying AECAs.
