Principle of Electrophoretic Mobility Shift Assay (EMSA)

EMSA is a biochemical procedure for characterizing protein-nucleic acid interaction (PNI). Nucleic acid fragments are often labeled for visualization purposes. In a classical assay, labeled nucleic acid and test protein are incubated, and the resulting mixture is subjected to electrophoresis. Since the molecular mass of the complex formed increases, the mobility of the complex decreases during this process. Since nucleic acid contains some special markers, the distribution of the substance can be determined by various methods, thereby proving the PNI. With the advancement of experimental technology, the assay can perform qualitative and quantitative analyses of the target.

EMSA is a popular in vitro method for detecting PNI. In addition to detecting transcription factors, this technique plays a crucial role in DNA-protein interactions, RNA-protein interactions and even DNA-RNA interactions.
