Mutations in EDA give rise to a clinical syndrome characterized by loss of hair, sweat glands, and teeth. EDA2R inhibits the proliferation and induces apoptosis of gastric cancer cells by regulating JNK signaling pathway.

BackgroundThe protein encoded by this gene is a type III transmembrane protein of the TNFR (tumor necrosis factor receptor) superfamily, and contains cysteine-rich repeats and a single transmembrane domain. This protein binds to the EDA-A2 isoform of ectodysplasin, which plays an important role in maintenance of hair and teeth. Alternatively spliced transcript variants encodes distinct protein isoforms.

Involvement in DiseaseDiseases associated with EDA2R include Ectodermal Dysplasia 1, Hypohidrotic, X-Linked and Ectodermal Dysplasia.
FunctionReceptor for EDA isoform A2, but not for EDA isoform A1. Mediates the activation of the NF-kappa-B and JNK pathways. Activation seems to be mediated by binding to TRAF3 and TRAF6.
