Exosome antibodies are antibodies developed against exosomal surface markers. Exosome Surface Markers

Exosomes, 30-150 nm in diameter, are endocytic membrane-derived vesicles that harbor a variety of proteins, nucleic acids and lipids, and are present in almost all bodily fluids. Many of the exosomal proteins are inserted in the membrane surface and some proteins that are most frequently identified are accepted as specific exosome markers, such as tetraspanins (e.g., CD9, CD63, and CD81 ), membrane transporters and fusion proteins (e.g., GTPases, annexins, and flotillin), and MVB biogenesis proteins (e.g., Alix and TSG101). Thus, antibodies against these markers can be developed for exosome studies. Moreover, some markers are disease-specific, making them potential biomarkers for disease diagnosis.

Immune Phage Display Antibody Library Screening

Constructing and screening immune antibody library via phage display technology has become a major choice for monoclonal antibody production. Unlike premade phage antibody libraries, animals are firstly immunized with exosome-associated antigens to produce antibody-secreting B cells and experience the in vivo affinity maturation process. This way, monoclonal antibodies with high specificity and affinity can be obtained more easily.

B Cell Sorting for Exosome Antibody Development

B cell sorting technology is a useful approach to develop antibodies for diagnostics, pharmacokinetic studies, and clinical development. This technology is based on first isolation of single B cells from a large population of primary B cells, followed by the direct amplification of VH and VL region encoding genes and their subsequent expression in cell culture systems.
