Fynomers Library Construction Service

Introduction of Fynomers

In recent years, the generation of binding molecules based on scaffold proteins have the huge potential to replace antibodies in the biological research and applications. Fyn is a 59 kDa member of the Src family of tyrosine kinases. In general, the Fyn SH3 can be composed with two antiparallel β-sheets and also contains two flexible loops which are RT loop and n-Src loop. These two loops play the important role in the protein interactions. Fynomers are small binding proteins (7 kDa) derived from the human SH3 domain of Fyn kinase which can be engineered to yield specific and high-affinity binding domains to target the specific proteins.

Construction of Fynomers Library

As for the Fynomers library construction, the gene encoding the Fyn SH3 domain can be amplified from the human cDNA library. With a series of researches, now Creative Biolabs has got the sequence of Fyn SH3 domain and the position sites of these two flexible loops. After the polymerase chain reaction with the corresponding primers, the resulting DNA fragment can then be inserted into the vector to form the Tyn SH3 WT. The most important stage of Fynomers library construction is the randomization of two flexible loops of the SH3 fold. In this case, the Tyn SH3 WT serves as the template and the PCRs are performed with the partially degenerate primers. And then, the randomized insert can be double-digested and cloned into the vector. After the electrical E. coli transformation, we can finally obtain the Fynomers library.
