Background: Effective vaccines inducing lifelong protection against many important infections such as the respiratory syncytial virus (RSV) are not yet available despite decades of research.
Results: B cells, engineered to express an antibody against RSV, gained the ability to bind RSV F antigen and resulted in potent and durable protection in RAG1-deficient mice with the phenotype consistent with long-lived antibody-secreting plasma cells and an isotype-switched memory B cell.
Q: Why functional assessment is needed?
A: Target antigen quantitation assessment could identify whether the B cells were correctly engineered and could play their basic anti-tumor role in vitro and in vivo. Phenotype and protection effect analysis is necessary to ensure that B cell immunotherapy could provide life-long protection in vivo.
