Protein drugs have been wildly used for the treatment of human diseases, including cancer, autoimmunity, and protein deficiencies. A major challenge in the development of protein drugs is poor pharmacokinetics and a short half-life. Conjugation of proteins with carriers, such as PEG2, Fc domains, albumin, or nanoparticles, can increase half-life, but these drugs still need recurrent delivery. A long-lived cell source is an alternative method to express and deliver protein drugs. Human long-lived plasma cell (PC) is a good source. PC is a terminally differentiated B cell population that resides in the bone marrow for decades and stably produces high concentrations of antibodies.

Hemophilia B is an X-linked bleeding disorder caused by a deficiency or dysfunction of FIX due to mutations in the FIX gene. Patients suffer from spontaneous hemorrhages and are classified as severe, moderate, or mild depending on their residual FIX activity levels. Hemophilia B is managed by intravenous injection of FIX protein, which is given prophylactically or on demand to manage bleeding episodes. The advent of B cell engineering technology has brought promise for the production of FIX protein in long-lived cell source-PCs.
Accumulated evidence has shown that therapeutic FIX protein can be delivered by antigen-specific B lymphocytes with significant levels in an antigen-regulated fashion in vivo. Our approach is suitable for the delivery of bioactive factors in chronic conditions. In the process, regulation is required over long periods but the rapidity and precise level of the response is not crucial.
