Target Antigen Quantitation Service

Introduction of Target Antigen Quantitation

Since B cells and plasma cells can support antitumor immune responses through several mechanisms, as shown in Fig.1. B cell engineering has been a valuable technology in the area of immunology therapy. Technically, B cells engineering is complicated and challenging, because they could bind their target antigen only after the engineering cassette is successfully inserted into the right locus. Even though, fully functional B cells still require post-translational modifications to produce membrane-bound and secreted antibodies. To fulfill this imperative need, Creative Biolabs offers the service of target antigen quantitation of B cells for the functional check.

Our Target Antigen Quantitation Service

We offer scientific and meticulous service for B cell target antigen quantitation, mainly including flow cytometry assay (FC), enzyme-linked immunosorbent assay (ELISA), and data analysis to ensure high-quality B cell engineering results. For more information about the assessment of engineered B cells functionally in vivo and in vitro, please inquire about our Engineered B Cells Assessment Service.

Published Data

Paper Title: B cells engineered to express pathogen-specific antibodies protect against infection

Technology: CRISPR/Cas9; Target Antigen Quantitation

Subjects: RAMOS B cells and Primary human B cells

Journal: Science Immunology

Published: 2019

Background: Effective vaccines inducing lifelong protection against respiratory syncytial virus (RSV) are not yet available despite decades of research.

Results: B cells engineered to express an antibody against RSV resulted in potent and durable protection in RAG1-deficient mice.

Frequently Asked Questions

Q: When the target antigen quantitation assay is needed?

A: Pre-experiments to test the feasibility of B cell engineering strategies; Evaluation of the strategy optimization effect of B cell engineering; Assessment of the antibody production level by engineered B cells in vitro and in vivo; Validation of the protective effect of animals from viral infection after transplantation of engineered B cells.

Q: What's the difference between FC and ELISA, two different target antigen quantitation assays?

A: FC could detect membrane-bound antibodies while ELISA could detect the secreted antibodies in the supernatant of cell culture or the serum of animals. Fully functional B cells could produce both membrane-bound and secreted antibodies.
